The 2nd Battalion fought the first major battles at Ap Bau Bang on 12 November 1965 and Ap Nha Mat on 5 December 1965. Heavy losses were suffered at Ap Nha Mat and three soldiers are still listed as missing. The 1st Battalion sustained its first major casualties of the war on 21 December 1965 when the enemy ambushed the command group of Company B as the company was moving out of Bien Hoa on routine patrol. On 25 August 1966 a patrol from Company C, 1st Battalion became involved in what became known as the Battle of Bong Trang with heavy losses on both sides.

Two battalions of 327th Infantry deployed to Vietnam in 1965, where they were joined by the rest of the division in late 1967. The 2nd Battalion, 327th Infantry, was the last unit of the 101st Airborne to leave Vietnam, departing in April 1972, making it the longest serving unit in combat during the Vietnam War. While still at Fort Campbell, Kentucky, the remaining elements of the 101st were severely stripped of personnel to support the war effort. When ordered to deploy, it was necessary to fill empty manning slots with non-airborne-qualified soldiers, effectively making these units of the division airborne in name only.

The Battalion was reactivated 12 August 1966 in Gulfport Mississippi as a Naval Mobile Construction Battalion. It seems the Battalion did not have a copy of the unit History from WWII with the Disney insignia and there was a belief that the Battalion's first deployment was supposed to have been Australia. This belief produced the Kangaroo insignia and the slogan "Kangaroo Can Do". After completion of training they deployed to Da Nang, Vietnam. The Battalion was awarded its second Navy Unit Commendation for this tour. The second deployment took them to Phu Bai Combat Base, Vietnam.
