The Battle of FSB Mary Ann occurred when Viet Cong (VC) sappers attacked the U.S. firebase located in Quảng Tín Province, South Vietnam early on the morning of 28 March 1971. On the early morning of 28 March sappers from the VC 409th Sapper Battalion approached the wire of FSB Mary Ann and took up positions to launch an attack. The exact number of sappers involved is uncertain, but most sources agree that there were at least 50. The battle at FSB Mary Ann inflicted serious losses on the defenders, who suffered 33 killed and 83 wounded.

From September through November 1966 the soldiers of "Tropic Lightning" participated in Operation Attleboro, East of Tay Ninh City, in War Zone C. 1966–1973: Approximately 5,000 US war dogs served in the Vietnam War (the US Army did not retain records prior to 1968); about 10,000 US servicemen served as dog-handlers during the war, and the K9 units are estimated to have saved over 10,000 human lives. 232 military working dogs and 295 US servicemen working as dog handlers were killed in action during the war. During the Vietnam War, the 1st Battalion, 46th Infantry and the 5th Battalion, 46th Infantry were part of the Americal Division. Both initially deployed to Vietnam with the 198th Infantry Brigade from Fort Hood, Texas. But the 1-46th was placed under operational control of the 196th Infantry Brigade (Light) and later became fully a part of the 196th Brigade. Though the 5-46th left Vietnam when the Americal Division cased its colors in the fall of 1971, the 1-46th deployed to Da Nang with the rest of the 196th. The 1-46th left Vietnam in June 1972.

On 7 October 1965, the 281st Assault Helicopter Company, Airmobile Light was activated at Fort Benning. By 9 June the following year, the newly formed company set up its headquarters inside the 5th SFG compound at Nha Trang Bay, Republic of Vietnam. The unit's mission was to provide air movement of troops, supplies and equipment under the direct supervision of the commanding officer, 5th SFG. The 281st became the first organized special operations helicopter unit in the U.S. Army. Army historians consider the 281st to be the legacy unit for today's 160th Special Operations Aviation Regiment (Airborne), abbreviated as 160th SOAR(A).

In all 53 members of the 16 SOS were killed in action during the Vietnam War.

Operation Junction City was an 82-day military operation conducted by United States and Republic of Vietnam (RVN or South Vietnam) forces begun on 22 February 1967 during the Vietnam War. It was the first U.S. combat airborne operation since the Korean War and one of the largest Airmobile operations of the war Junction City's grand tactical plan was a "hammer and anvil" tactic, with airborne forces "flushing out" the VC headquarters, driving it against a prepared "anvil" of other forces. As was common practice for such units, the sappers wore khaki shorts and soot camouflage and were armed with either an AK-47 or RPG-7 and satchel charges and grenades to attack bunkers. Sappers relied on stealth, shock and surprise to give them an advantage, and rarely carried heavy weapons or equipment.

The squadron was reactivated as the 430th Fighter-Bomber Squadron in 1952, when it replaced an Air National Guard squadron in Japan. It again saw combat in the Korean War, earning another Distinguished Unit Citation and a Republic of Korea Presidential Unit Citation. The squadron returned to the United States in 1954, but made frequent overseas deployments, including combat in the Vietnam War. It remained a fighter unit until inactivating in 1989. In 1992, the unit became the 430th Electronic Combat Squadron, but was only active for one year. It was converted to provisional status and received its current name in 2013.

Kain, Jr. (radar intercept officer), in an F-4J Phantom from VF-33 downed a MiG-21 'Fishbed', 17 mi (27 km) northwest of Vinh, North Vietnam, for the ship's first MiG "kill" in the Vietnam War. America and her embarked air wing, CVW-6, would later be awarded the Navy Unit Commendation for their work during that time.
